Custom antibody development

We offer a customized service for the identification of high-affinity single domain antibodies (sdAb) targeting membrane proteins, using phage display technology applied to recombinant extracellular vesicles (rEVs). This approach allows selection of sdAb against membrane proteins in their native conformation, preserving structural integrity and functional epitopes.

Applications

  • Therapeutic antibody discovery
  • Diagnostic development
  • Target validation
  • Antibody-based imaging or delivery tools

Our Workflow

  1. Design and Production of rEVs expressing the membrane protein of interest (GPCR, CD like antigen, channel, transporter…).
  2. Characterization of rEVs (Western blot, biolayer interferometry, electron microscopy)
  3. Llama immunization and VHH library construction
  4. Phage display screening: multiple rounds of biopanning on rEVs
  5. Clone screening (ELISA)
  6. Sequencing and analysis of enriched antibody fragments
  7. Affinity and specificity validation
  8. Optional sdAb modifications (eg Fc), affinity constant determination (KD), structural analysis

Advantages

  • Preservation of native structure of the membrane protein in lipid context
  • Increased specificity by eliminating irrelevant binders during biopanning by using Mock rEVs
  • Confidential and modular service tailored to your project
  • Suitable for rare or low-abundance targets

Timeline & Deliverables

  • Project duration: 5 – 6 months
  • Deliverables: Sequencing data, binding analysis report, optional expression vectors or purified antibodies
