Protein structure determination by Cryo-EM

Our Cryo-EM protein analysis service, powered by the Thermo Scientific™ Glacios™ Cryo-TEM (200 kV), offers high-throughput and high-quality structural insights for proteins and complexes in near-native states. This platform is ideal for screening and resolving protein structures with resolutions typically ranging from 3 to 5 Å, with potential for near-atomic resolution depending on sample quality.

Service Overview

We provide a complete Cryo-EM pipeline using the Glacios™ system, including:

  • Sample Preparation & Vitrification
    Vitrification of purified protein samples using an automated Thermo Vitrobot™ or Leica GP systems for reproducible grid freezing under controlled humidity and temperature.
  • Cryo-Grid Screening
    Evaluation of multiple cryo-grids to assess ice thickness, particle distribution, and sample behavior directly on the Glacios microscope.
  • High-Throughput Data Collection
    Automated data acquisition using direct electron detection (Falcon™ 4 camera or equivalent), with optimized dose fractionation and defocus ranges.
  • Image Processing & 3D Reconstruction
    Processing via industry-standard software suites such as CryoSPARC™, RELION for particle picking, 2D/3D classification, and refinement.
  • Structural Modeling (Optional)
    Density map interpretation, rigid-body fitting, and model refinement using tools like Phenix, ChimeraX, or Rosetta.

Deliverables

  • Cryo-EM raw movie stacks and aligned micrographs
  • 2D class averages and 3D density maps
  • Resolution estimation and FSC validation curves
  • Optional fitted atomic model (if resolution allows)
  • Full analysis report summarizing workflow and results

Sample Requirements

  • Protein purity: ≥90%
  • Concentration: 0.5–5 mg/mL
  • Volume: 50–100 µL minimum per condition
  • Buffer: Cryo-compatible, low in glycerol and detergents
  • Particle size: Ideally >100 kDa for optimal visualization on Glacios
